Two-photon, fiber-coupled, super-resolution microscope for biological imaging

چکیده

Imaging sub-diffraction dynamics of neural nanostructures involved in behaviors such as learning and memory a freely moving animal is not possible with existing techniques. Here, we present solution the form two-photon (2P), fiber-coupled, stimulated emission depletion microscope demonstrate its capabilities by acquiring super-resolution imaging mammalian cells. A polarization-maintaining fiber used to transport both 2P excitation light (915 nm) donut-shaped beam (592 nm), which constructed adding two temporally incoherent orthogonally polarized Hermite–Gaussian modes. The output insensitive bending or temperature changes first demonstration toward deep tissue awake behaving animals.
